STUDY ON THE HPLC DETERMINATION OF POLYAMINES IN PERSIMMON

摘要 采用高效液相色谱法,用1,6 己二胺为内标,定量测定柿子中的精胺(Spermine)、亚精胺(Spermidine)、尸胺(Cadaverine)和腐胺(Putrescine)。样品经丹磺酰氯衍生,硅胶柱净化,LichrosorbNH2柱分离后,以氯仿∶乙酸乙酯∶三乙胺(98.4∶1.5∶0.1)为流动相,流速为1 5ml·min-1,荧光检测器检测。激发波长为336nm,发射波长为495nm,精胺、亚精胺、尸胺和腐胺的线性相关系数均大于0.99,其回收率分别为98.99%、94.02%、96.46%和105.44%。 Polyamines, including spermine (SPM), spermidine (SPD), cadaverine (CAD), and putrescine (PUT) in persimmon were separated by HPLC, after DNS-Cl derivatization, on a Lichrosorb-NH_2 column (5μm, 200mm×4.0mm), using a mixed solution composed of chloroform, ethyl ether and triethylamine in a ratio of 98.4 to 1.5 and to 0.1 correspondingly, as the mobile phase with a flow rate of 1.5ml·min^(-1). Fluorimetric detector was used in the final determination with λ_(ex)=366nm and λ_(em)=495nm. Average recoveries found were 98.99% (for spermidine), 94.02% (for spermine), 96.46% (for cadaverine) and 105.44% (for putrescine).
