摘要
目的 验证新生大鼠骨髓基质细胞的成骨特性及扩增能力 ,从而为今后的临床工作提供一定的基本理论基础。方法 分离新生 7天Wistar大鼠股骨及胫骨 ,冲洗骨髓腔并培养 ,用成骨细胞诱导液培养诱导 9~ 1 2天 ,分别用HE染色及骨碱性磷酸酶 (BALP)染色 ,通过对阳性BALP活性分析确定BMSC的成骨特性。结果 经过 9~ 1 2天诱导后骨髓基质细胞的形态为成骨细胞样BALP活性明显增强。结论 新生大鼠骨髓基质细胞体外扩增能力强 。
Objective To verify the osteogenic property and the expanded ability of bone marrow stem cells in neonatal rats.Methods Thighbone and shankbone in neonatal rats were separated,and their marrow cavity were washed.The bone marrow stem cell was cultivated by osteoblast induced liquor for 9~12 days.Then,it was dyed by HE and BALP.By analyzing the living BALP in bone marrow stem cell,we could conclude that bone marrow stem cell might differentiate into osteoblast,and could be cultivated outside of body stably.Results Bone marrow stem cell could be induced into osteoblast and the living BALP in BMSC increased in vitro. Conclusion The bone marrow stem cell in newborn rats has strong ability to expand and to differentiate into osteoblast visibly.
