摘要
目的 研究血糖安对 2型糖尿病大鼠丙酮酸激酶活性的影响。方法 将大鼠灌胃脂肪乳 1 0天后应用四氧嘧啶建立 2型糖尿病模型。动物分为正常组 ,高脂组 ,糖尿病组 ,苯乙双胍组 ,血糖安大、中、小剂量组。灌胃 7天后 ,测定各组血糖值 (血糖仪 )、肝脏和血清中丙酮酸激酶活性 (比色法 )。结果 ①血糖安降低 2型糖尿病大鼠血糖值 ,大、中、小剂量从治疗前 (1 9.98± 3.89)mmol L、(2 0 .88± 4 .1 9)mmol L、(2 1 .1 2± 5 .1 4 )mmol L至治疗后 (8.0 3±2 .89)mmol L、(7.6 8± 3.2 7)mmol L、(1 0 .2 7± 3.4 2 )mmol L (P <0 .0 1 ) ;②血糖安升高 2型糖尿病大鼠的丙酮酸激酶活性 ,在肝脏中 ,中、大剂量由糖尿病时的 (4 2 6 .2 3± 2 6 .2 0 )U L增加到 (4 4 9.5 1± 9.2 6 )U L ,(4 5 4 .99± 1 1 .95 )U L(P <0 .0 5 ) ;在血清中由 (1 81 .2 1± 4 2 .73)U L增加到 (2 6 5 .6 1± 5 3.5 1 )U L ,(30 1 .71± 5 8.1 4 )U L(P <0 .0 5 )。而且大剂量在肝脏中 ,中、大剂量在血清中使酶活性完全恢复 (P >0 .0 5vs正常组 )。结论 血糖安降低 2型糖尿病大鼠血糖值 。
Objective To study the effects of Xue Tang An(XTA) on pyruvate kinase activity in type 2 diabetic rats.Methods Rats were fed with high fat food for ten days to grow into obesity,alloxan was injected ip to make type 2 diabetic model.Rats were divided into normal group,high fat group,diabetic model group,phenformin group,and XTA group (high,medial and low dose).Continually treating them for seven days,then measuring the blood glucose level (blood glucose meter) and pyruvate kinase activity (colorimetry).Results ①XTA decreased the blood glucose level from pre treatment(19.98±3.89)mmol/L,(20.88±4.19)mmol/L,(21.12±5.14)mmol/L to post treatment(8.03±2.89)mmol/L,(7.68±3.27)mmol/L,(10.27±3.42)mmol/L ( P <0.01)with high,medial and low dose,respectively.②XTA increased pyruvate kinase activity of type 2 diabetic rats from(426.23±26.20)U/L in diabetes to(449.51±9.26)U/L,(454.99±11.95)U/L ( P <0.05) in liver and from(181.21±42.73)U/L to (265.61±53.51)U/L,(301.71±58.14)U/L ( P <0.05) in serum with medial and high dose,respectively.Pyruvate kinase activity was restored ( P >0.05 vs normal group) with high dose in liver,medial and high dose in serum.Conclusion XTA decreases the blood glucose level of type 2 diabetic rats through improving the activity of pyruvate kinase which may be a key chain of its effects.
