摘要
目的 观察腺苷A1受体激动剂 2 氯化腺苷 (2 CAdo)对马桑内酯致痫大鼠海马神经细胞bcl 2 ,Bax基因表达的影响。方法 采用马桑内酯诱导癫痫发作模型 ,通过免疫组织化学方法检测bcl 2 ,Bax蛋白来观察大鼠海马CA1区神经细胞凋亡的动态变化及 2 CAdo对其的影响。结果 海马CA1区癫痫发作后 2 4hbcl 2表达量增多 ,4 8h明显下降 ,72h仅有少量表达 ,7天时其表达量再度升高。而Bax表达则在癫痫发作 2 4h开始增多 ,4 8h显著增多 ,72h表达达高峰 ,之后逐渐减少 ,7天表达量最少。给予 2 CAdo组各相应时间点bcl 2表达量较对照组明显增高 (P<0 .0 5 ) ,Bax表达量较对照组明显减少 (P <0 .0 5 ) ,经统计学处理均有显著性差异。结论 2 CAdo减少癫痫发作后海马神经细胞的凋亡 。
Objective To investigate the influence of 2 CAdo(which is the activation of A1 receptor) on the genes bcl 2 and Bax expression of the neuron in the hippocampus of the rats induced epilepsy by coniaria lactone(CL).Methods In the model of coniaria lactone induced epilepsy,the amount of apoptotic neuron in the hippocampus CA1 area was measured continually and influence of 2 CAdo both genes was observed by detecting bcl 2,Bax protein with immunohistochemical method.Results The expression of bcl 2 in CA1 was increased at 24 h after seizures,significantly decreased at 48 h,even less at 72 h,and the expression of bcl 2 was increased at 7 day again; whereas Bax was increased at 24 h after seizures,significantly increased at 48 h.The peak level of Bax expression was reached at 72 h,then decreased,lowest at 7 day.After given 2 CAdo,the expression of bcl 2 was significantly higher than that of normal control group at the corresponding time ( P < 0.05 ),the expression of Bax was significantly lower than that of normal control group ( P <0.05).Conclusion 2 CAdo might decrease the amount of apoptotic neuron in rats hippocampus after induced seizures,and protect neurons.
