白血病诱导分化治疗的新策略:组蛋白乙酰化/去乙酰化

摘要 核心组蛋白含带正电荷的碱性氨基酸,与DNA具有高度亲和力,其乙酰化状态与基因的转录激活有关,组蛋白去乙酰化酶抑制剂(HDACIs)能调节组蛋白的乙酰化状态,在基因表达调控中发挥着重要作用,通过抑制组蛋白去乙酰化酶(HDAC)活性提高组蛋白乙酰化水平,可以促进细胞分化,HDACIs能诱导白血病和对维甲酸耐药白血病的分化,在体外及体内动物模型均得到证实,其作用机制完全不同于维甲酸,有些HDACIs已进入临床一、二期实验,具有较好的疗效,毒副作用小,展现出良好的应用前景,目前已成为白血病诱导分化治疗的一条新途径。
