甲基化修饰对宫颈癌细胞凋亡相关蛋白激酶表达的调控

摘要 目的 :探讨在 DNA甲基转移酶抑制剂 5 -氮胞苷 ( 5 - azacytidine)的化学干预下 ,宫颈鳞癌细胞 Si Ha和宫颈腺癌细胞 He La中凋亡相关蛋白激酶 ( DAPK)基因与甲基化调控的关系。方法 :培养 Si Ha、He L a,以 1 0μmol/L浓度的 5 -氮胞苷干预细胞。提取细胞RNA,用逆转录聚合酶链反应 ( RT- PCR)方法检测凋亡相关蛋白激酶基因 m RNA的表达。结果 :He La细胞的 DAPK基因用 5 -氮胞苷干预前后无明显变化。 Si Ha中 DAPK基因表达缺失 ,用 5 -氮胞苷处理后 ,Si Ha中 DAPK基因可重新表达。结论 :在不同类型宫颈癌细胞中 DAPK基因的调控有明显差异。 DAPK基因在宫颈鳞癌细胞中的表达受 DNA甲基化调控 。 Objetive: To investigate the effects of DNA methylation on tumor suppressor genes death associated protein kinase(DAPK) in human cervical cancer cell lines by DNA methyltransferase inhibitor, 5 azacytidine (5 aza) intervention. Methods: cancer cell lines SiHa?HeLa were treated with 5 aza in 10μmol/L concentrations. The expressions of genes DAPK were assayed by reverse transcriptase polymerase chain reaction (RT PCR). Results:Expression of tumor suppressor gene DAPK was detected in HeLa cells and transcription levels were not changed after treatment. DAPK expression was not expressed in SiHa cells and was reexpressed after 5 aza treatment. Conclusions: The effects of DNA methylation on of DAPK in different human cervical cancer cell lines are different. The expression of DAPK can be regulated by DNA methylation in SiHa cells, but not in HeLa cells.
