胃癌中cyclinD_2和p27^(kipl)的表达及其意义

Significance of cyclinD_2 and p27^(kipl) Expressed in Gastric Carcinoma

目的 探讨cyclinD2 、p2 7kipl的表达与胃癌发生发展的关系。方法 用免疫组化方法检测cy clinD2 和p2 7kipl在 12 0例胃癌、2 0例胃粘膜非典型增生、3 0例正常胃粘膜中的表达。结果 cyclinD2 在正常胃粘膜、非典型增生、胃癌中阳性表达率分别是 3 .3 %、10 %、和 3 4 .2 % ,胃癌与正常胃粘膜及非典型增生相比有显著性差异 (P <0 .0 1) ,p2 7kipl在正常胃粘膜、非典型增生、胃癌中表达分别是 10 0 %、90 %、和 52 .5%。胃癌与其他两组比较有显著性差异 (P <0 .0 1) ,p2 7kipl表达缺失和cyclinD2 胞浆阳性者与胃癌的分化程度、浸润深度、淋巴结转移有关 (P <0 .0 1)。cyclinD2 和p2 7kipl表达呈负相关。结论 cy clinD2 和 p2 7kipl与胃癌的发生有密切关系 ,p2 7kipl的表达缺失及cyclinD2 胞浆阳性者与胃癌的进展有关 ,p2

Objective To investigate the relationship between the expression of cyclinD 2 and p27 kipl in carcinogenesis and development of gastric carcinoma. Methods cyclinD 2 and p27 kipl were detected by S P immunohistochemical method in 120 cases of gastric carcinoma, 20 cases of dysplastic gastric mucosa and 30 cases of normal gastric mucosa. Results The positive expression rate of cyclinD 2 in cases with gastric carcinoma?dysplasia and normal gastric mucosa were 3.3% , 10% and 34.2% respectively. There was significant difference between carcinoma and normal gastric mucosa or dysplasia( P < 0.01 )?The positive expression rate of p27 kipl in cases with carcinoma, dysplasia and normal gastric mucosa were 52.5% , 90% and 100% respectively. There was significant difference between carcinoma and normal gastric mucosa or dysplasia( P < 0.01 )? The lack of p27 kipl and cyclinD 2 cytoplasmic staining correlated with differentiation and depth of invasion and lymph node metastasis of gastric carcinoma ( P < 0.01 )?There was an inverse relationship between cyclinD 2 and p27 kipl expression. Conclusion The expression of cyclinD 2 and p27 kipl may play important role in the gastric carcinogenesis. The loss of p27 kipl protein expression and cyclinD 2 cytoplasmic localization are related to progression of gasric carcinoma and p27 kipl might be a useful prognostic marker in gastric carcinoma .
