摘要
目的 探讨Oncolyn对温石棉所致肺泡巨噬细胞氧化损伤的影响。方法 应用支气管灌洗的方法获得大鼠肺泡巨噬细胞 (AM) ,纯化后与终浓度 5 0 μg/ml的温石棉分别孵育 2h和 4h ,在孵育的同时加入不同浓度的Oncolyn ,检测AM中DNA损伤状况以及抗氧化酶SOD、GSH Px的活性。结果 肺泡巨噬细胞与温石棉共同培养 2h后 ,DNA损伤和SOD、GSH Px活性增加 ,表现出彗星出现率和DNA迁移长度增加 ;当培养 4h后 ,彗星出现率比培养 2h更高 ,DNA迁移长度不再增加 ,而SOD、GSH Px活性均下降。在AM与温石棉作用的同时加入不同浓度的Oncolyn后 ,DNA损伤减轻 ,抗氧化酶SOD、GSH Px的活性上升。
Objective To explore the effect of Oncolyn on oxidative damage in alveolar macrophages(AM)induced by chrysotile.Method AMs of rats were collected by bronchial lavage and cultured with 50 μg/ml chrysotile and Oncolyn for 2 h or 4 h,then detecting the damage status of DNA and the activities of SOD and GSH-Px of cultured AMs.Result After 2 h culturing with chrysotile the comet cell incidence,the DNA migration length and the activities of SOD and GSH-Px of AM were all significantly increased.When AMs cultured with chrysotile for 4 h,the damage level of DNA was continually increased,while the activities of SOD and GSH-Px showed a obvious decrease.The addition of Oncolyn with chrysotile simultaneously made the comet cell incidence,the DNA migration length decreased,and the activities of SOD and GSH-Px increased.Conclusion It is suggested that Oncolyn has some protection or remission effect on oxidative damage of AM caused by chrysotile.
