摘要
目的 探讨复方丹参注射液球后注射对视网膜缺血再灌注 (retinalischemiareper fusion ,RIR)损伤的防护作用及机制。方法 采用前房灌注液体形成 14 .3kPa高眼压而建立RIR模型。在损伤前 30min给予防护组大鼠球后注射复方丹参注射液 0 .0 5mL ,对照组大鼠球后注射生理盐水 0 .0 5mL ,缺血 6 0min后恢复血流 ,分别于再灌注 30min ,2 4h和 72h检测视网膜组织中丙二醛 (MDA)含量及视网膜电图 (ERG)中b波变化 ,并进行视网膜透射电子显微镜和光学显微镜的组织学观察。结果 再灌注 30min ,2 4h和 72h后防护组视网膜中MDA含量均明显低于对照组 (P <0 .0 1) ,而ERG中被标化的b波振幅均高于对照组 (P <0 .0 1) ,且防护组病理组织学损害较对照组明显减轻。结论 复方丹参注射液球后注射是防护RIR损伤的有效方法。
Objective It is to investigate the protective effect and mechanism of salvia miltiorrhiza composite retrobulbar injection from retinal ischemia reperfusion-induced retinopathy. Methods Retinal ischemia was induced in SD rats by increasing the intraocular pressure to 14.3*!kPa for 60 minutes via cannulation into the anterior chamber. 30 minutes before retinal ischemia, the rats in the protective group received a retrobulbar injection of salvia miltiorrhiza composite retrobulbar injection at a dose of 0 05*!mL and the rats in the control group received a retrobulbar injection of sodium chloride at a dose of 0 05*!mL. At 30 minutes, 24 hours and 72 hours after ischemia reperfusion, the content of malandialdehyde (MDA) in rat retinas were measured, the changes of b-wave in electroretinography (ERG) were observed and histopathologic changes were observed with transmission electron microscopy and light microscopy. Results At 30 minutes, 24 hours and 72 hours after ischemia reperfusion, the content of MDA in rat retinas in protective group significantly less than that in control group (P<0 01), while the normalized b-wave amplitudes in the protective group were significantly higher than that in the control group (P<0.01). The histopathologic injury of retina in the control group was less than that in the protective group. Conclusion Retrobulbar injection of salvia miltiorrhiza composite retrobulbar injection is an effectively protection method for retinal ischemia reperfusion-induced retinopathy.
