摘要
目的 :观察“芪箭消瘿汤”对实验性自身免疫性甲状腺炎小鼠自身抗体、甲状腺病理、甲状腺表皮生长因子受体 ( EGFR)表达的作用及机制。方法 :应用人甲状腺球蛋白与完全弗氏佐剂混合免疫注射法模拟小鼠实验性自身免疫性甲状腺炎 ,用被动血凝法检测甲状腺自身抗体水平 ,HE染色观察小鼠甲状腺组织病理改变 ,S- P免疫组化染色观察 EGFR表达。结果 :芪箭消瘿汤明显抑制甲状腺自身抗体水平 ,减轻小鼠甲状腺组织淋巴细胞浸润 ;模型组小鼠甲状腺 EGFR表达高于正常对照组 ( P<0 .0 5 ) ,芪箭消瘿汤大剂量组低于模型组 ( P<0 .0 5 ) ,芪箭消瘿汤小剂量组、雷公藤片组与模型组比较差异无显著性意义 ,P>0 .0 5。结论 :芪箭消瘿汤可调节桥本氏甲状腺炎的自身免疫紊乱 ,减轻 EGFR的过度表达 ,对小鼠实验性自身免疫性甲状腺炎疗效明显。
Objective: To observe the effect of Qijianxiaoying decoction on the auto-antibody, pathology of thyroid and expression of epidermic growth factor recptor(EGFR) in experimental autoimmune thyroiditis(EAT) mice, and to study its mechanism. Methods:The EAT model was induced by injecting hTg and CEA. The level of auto-antibody was checked by PHA. The pathology change of thyroid tissue was observed through HE colouration. And the expression of EGFR was investigated by S-P immune histochemistry.Results: Qijianxiaoying decoction could significantly suppress the level of auto-antibody and reduce lymphocyte soakage of murine thyroid tissues. The expression of EGFR in model group was significantly higher than that in normal control group( P <0.05), that in great dose group being lower than in model group( P <0.05), while being no significant difference among small dose group, Leigonten tab group and model group( P >0.05). Conclusion:Qijianxiaoying decoction can adjust the auto-immune disorder in Hashimoto's thyroiditis, decline the excessive expression of EGFR, which have a good curative effect on EAT.
