调心方对氧化损伤型类AD大鼠脑组织淀粉样蛋白(APP) mRNA表达的影响 被引量:2

摘要 目的 :观察调心方对活性氧所致的氧化损伤型类AD大鼠脑组织NF κB转录因子和APPmRNA水平的影响。方法 :用二羟延胡索酸 (DHF)加FeCl3 ADP左侧脑室注射法复制氧化损伤型类AD大鼠模型 ;Morris水迷宫法检测AD大鼠空间记忆能力 ;RT PCR法检测鼠脑皮层和海马APPmRNA水平 ;免疫组化法检测鼠脑皮层和海马NF κB转录因子表达 ,以及调心方对上述各项指标变化的影响。结果 :与正常对照组比较 ,AD模型鼠空间记忆能力显著下降 ,脑组织NF κB转录因子和APPmRNA水平明显增高 ;调心方对上述指标有明显的改善作用。结论 :调心方能够通过下调AD大鼠脑组织NF κB转录因子和APPmRNA表达水平 ,减少Aβ产生 ,改善AD大鼠的空间记忆能力。 Objective: To observe the effect of the Heart Benefiting Recipe (HBR,调心方) on the NF-κB transcription factor and APP mRNA level in cortex and hippocampus of the oxidative damaged AD rat model. Methods: The rats were injected with the oxygen free radical generation system (DHF-FeCl_3-ADP) in left ventricles of the brain and induced oxidative damaged AD rat model. Using Morris water maze method to determine the changes of rats space memory. The level of APP mRNA (RT-PCR) and the expression of NF-κB (immunohistochemical method) were also observed. The regulating effects of HBR on above indices were also studied. Results: Compared with the normal group, the level of APP mRNA and NF-κB transcription factor in AD rat model were increased obviously. HBR, however, could decrease that of the AD rat model significantly. Conclusion: HBR can reduce the produce of Aβ through decreasing the level of APP mRNA and NF-κB transcription factor so that improves the space memory.
