摘要
目的 通过PCR -SSP法检测羊水细胞A1、2 、BO1、2 血型基因型 ,产前诊断胎儿ABO血型。方法 用PCR -SSP检测 5 6例孕 16w以上孕妇羊水细胞ABO血型基因型、用常规方法检测双亲及其胎儿或新生儿脐血ABO血型。结果 (1) 5 6例羊水细胞用PCR -SSP法均检测出ABO血型基因型及其分型 ;(2 )用常规血型检测方法检测脐血红细胞ABO血型抗原检出率为 87 5 % ;(3)PCR -SSP法检测出羊水细胞ABO血型与父母血型按孟德尔遗传规则测出的胎儿血型符合率为 10 0 %。结论 (1)PCR -SSP法检测羊水细胞ABO血型基因型是准确产前诊断胎儿血型的方法。 (2 )PCR -SSP法检测羊水细胞ABO血型基因型可以精确至其亚型。
Objective:Detecting amniotic fluid cell A 1、2,BO 1、2 genotype by PCR-SSP to accurately diagnose fetal ABO blood group.Methods:The subjects were fifty eight pregnant women with gestational age≥16 weeks, their newborns (or fetus) and their husbands.All amniotic fluid cells and fetal or newborns's umbilical blood were detected by polymerase chain reaction with sequence-specific primers.The parental and umbilical blood erythocytes group was detected by general method.Results: (1) ABO blood group genotypes of amniotic fluid cell were detected by PCR-SSP in fifty eight pregnant women.(2) Detected rate of fetal or newbrons's blood group by general method is 87.5%.(3) dentical rate of blood group of amniotic fluid cells with parental genetic blood group is 100%. Conclusion:(1) accurate prenatal diagnosing method.(2) The way of testing fetal ABO blood group by PCR-SSP with amniotic fluid cell can divide into the ABO blood subgroup.
