摘要
目的 研究COX - 2和iNOS在大肠腺癌中的表达和相互作用 ,其及它们与大肠腺癌生物学行为的关系。方法 采用SP免疫组化方法检测了 76例手术切除的大肠腺癌标本和 15例正常肠粘膜内COX - 2和iNOS蛋白的表达。结果 COX - 2和iNOS在大肠腺癌中阳性率 (76 .3%、85 .5 % )显著高于正常大肠粘膜(13.3%、2 0 % ,P <0 .0 0 5 ) ;不同组织类型大肠腺癌之间COX - 2阳性积分有显著差异 (P <0 .0 0 5 ) ,乳头状腺癌、管状腺癌Ⅰ级和管状腺癌Ⅱ级显著高于管状腺癌Ⅲ级 (P <0 .0 0 5 )和粘液腺癌 (P <0 .0 0 5 ) ;有淋巴结转移和无淋巴结转移者COX - 2的表达无显著差异 (P >0 .0 5 ) ;不同组织类型大肠腺癌之间iNOS阳性积分无显著差异 (P >0 .0 5 ) ;有淋巴结转移组iNOS的表达显著高于无淋巴结转移组 (P <0 .0 1) ;大肠腺癌组织中COX- 2和NOS的表达无相关性 (χ2 =0 .15 35 ;P >0 .0 5 )。结论 COX - 2和iNOS过度表达参与了大肠腺癌的发生展过程 ,并与其生物学行为有关 ;在大肠腺癌中COX - 2和iNOS的表达无相关性 ,提示它们在大肠腺癌的发生发展中所起的作用不同 ,是两个相互独立的作用因子。
Objective:To study the expression and interaction of COX-2 and iNOS in colorectal adenocarcinoma and the relationship between both COX-2 and iNOS expression and biology behavior of colorectal adenocarcinom.Methods:76 intestinal biopsy specimens were taken from patients with colorectal adenocarcinoma and 15 normal subjects for controls(NS), the expression of COX-2 and iNOS proteins in intestinal mucosal cells was detected by immunohistochemical staining(SP method) in the 96 cases .Results:In colorectal adenocarcinom the expression of COX-2 and iNOS protien was significantly higher than in NS (P<0.005) . The expression of COX-2 protein was significantly higher in papillary adenocarcinoma, tubular adenocarcinoma grade Ⅰ and grade Ⅱ than in tubular adenocarcinoma grade Ⅲ and mucinous adenocarcinoma(P<0.005).There was not significant difference between patients with metastasis and nonmetastasis for expression of COX-2(P>0.05).There was no significant difference among different pathological types for expression of iNOS(P>0.05).The expression of iNOS protein was significantly higher in patients with lymph node metastasis than without metastasis(P<0.01).There were no correlation between the expression of COX-2 and iNOS protein (χ2=0.1535; P>0.05) .Conclusions:The excessive expression of COX-2 and iNOS participates in genesis of colorectal carcinoma and was concerned with the biological behavior of colorectal adenocarcinom.The COX-2 and iNOS participate in genesis of colorectal carcinomas independent of each other.
