摘要
目的 探讨大鼠肝癌中组胺H1受体基因的mRNA表达情况。方法 采用二甲基氨基偶氮苯 (DAB)诱发大鼠肝癌模型 ,以逆转录聚合酶链反应 (RT PCR)半定量法研究肝癌组织、远癌肝组织与正常肝组织中组胺H1受体基因的mRNA表达 ,并将其PCR产物进行测序。结果 组胺H1受体mRNA在肝癌组织中的表达明显低于远癌肝组织 (P <0 0 1)和对照组肝组织 (P <0 0 1) ,且肝癌中组胺H1受体mRNA的碱基发生点突变。结论 组胺H1受体基因的转录减少以及H1受体基因的点突变可能与DAB诱导的大鼠肝癌形成有关。
AIM To study the mRNA expression of histamine H_1 receptor in hepatocarcinoma of rats. METHODS Dimethylamino-azobenzene (DAB) was used to induce hepatocarcinoma in rats. Semi-quantitative reverse transcription-polymerase chain reaction (RT-PCR) was adopted to analyse the relative expression of histamine H_1 receptor. And the base sequence of its PCR product was detected. RESULTS The relative mRNA expression of histamine H_1 receptor was significantly decreased in hepatic carcinoma tissue, compared with that part far from cancer and control group (P<0.01, respectively). And gene mutation was detected in the base sequence of H_1 receptor mRNA in hepatic carcinoma tissue. CONCLUSION Decrease in mRNA expression of histamine H_1, receptor and point mutation to H_1 receptor gene may play a role in the occurrence and development of rats' hepatocarcinoma induced by DAB.-
