摘要
目的建立一种测定硫酸软骨素含量的新方法。方法以乳化剂OP作稳定剂 ,在pH 3.0的磷酸 磷酸二氢钾缓冲液中碱性艳蓝BO与硫酸软骨素反应形成离子对缔合物 ,水溶液颜色由绿色变为蓝色 ,分光光度法测定硫酸软骨素含量。结果在 5 70nm波长处 ,碱性艳蓝BO与硫酸软骨素缔合物的吸收系数为 933。线性范围 0~ 4 .0μg/ml,RSD为 2 .2 %。 结论此方法简便、快速。
PurposeTo establish a new method for the determination of chondroitin sulfate (CS).MethodsIn the presence of emulsifier OP and H 3PO 4-KH 2PO 4 buffer solution, Victoria Pure Blue BO reacted with CS forming associated complex.The colour of the solution changed from green to pure blue. A method of spectrophotometric determination for CS was developed.ResultsThe maximal absorption of the solution was at 570 nm and the apparent absorptivity of associated complex of Victora Blue BO with chondroitin was 933. The absorption of CS solution obeyed Beer′s law, the concentration range was 0 to 4.0 μg/ml, and RSD was 2.2%.ConclusionThe method is accurate,simple and quick.
