摘要
目的 :对 15号染色体上控制小鼠脊髓颈膨大截面积的数量性状基因座 (QTL)进行精细定位。方法 :以高级互交系小鼠 (A/J)X(C5 7BL/6J) (F4)为研究对象 ,在D15Mit15 8位点附近作高密度图谱的基因组扫描 ,用MapManagerQTX13对脊髓颈膨大截面积与基因型进行连锁分析。结果 :在 15号染色体D15Mit10 7和D15Mit2 0 8附近出现了两个连锁峰 ,LRS值分别为 7.2 (P <0 .0 5 )和 6.4(P <0 .0 5 )。结论 :原 15号染色体上控制脊髓颈膨大截面积的一个QTL实际包含有两个QTL ,分别位于D15Mit10 7、D15Mit2 0 8附近 ,宽约 3 .2cM和 10 .
Objective:To fine map quantitative trait loci controlling the cross section area of cervical enlargement on chromosome 15 of mice. Methods:An advanced intercross line (AIL)(F4) was used for genome scanning. High-resolution microsatellite markers located within the previous QTL,which was detected on chromosome 15 , were genotyped on the mouse population representing phenotypic extremes. Linkage analysis was performed by computer software Map Manager QTX13.Results: Linkage sites were identified around D15Mit107 (LRS=7.2, P<0.05) and D15Mit208 (LRS=6.4, P<0.05) respectively. Conclusions:The former QTL on chromosome 15 actually comprised two QTLs, which were fine mapped respectively to an interval of 3.2cM and 10.2 cM.
