摘要
目的 探讨人前列腺干细胞的分离方法并初步鉴定其特性。方法 以器官捐献者的正常前列腺为研究对象 ,利用免疫磁珠细胞分离和胶原快速粘附方法 ,从前列腺基底细胞中分离前列腺干细胞 ,研究其克隆形成和分化特性。结果 从正常人前列腺内分离得到的细胞 ,能快速粘附胶原 ,克隆形成率明显高于未粘附的前列腺基底细胞 ,而且形成的克隆维持时间较长。结论 人前列腺基底细胞中存在极少量的具有高度增殖能力的一群细胞 ,它们具有干细胞的特征 ,但有待于进一步研究鉴定。
Objective To explore the methods of isolating the normal human protate stem cells and to identify its characteristics. Methods Using immunologic magnetic beads and the characteristics of stem cells fast adhering to collagen, clonal analysis of normal human prostate putative stem cells from basal cells were undertaken to identify stem cells.Results A small group of cells isolated from prostatic basal cells were characterized by fast adhesion to collagen I and high clone forming efficiency(CFE),which was much more high than that of the homogenetic prostatic basal cells.Conclusion On the basis of their high proliferative characteristics,the group of cells may be prostate stem cells or their progeny.
