摘要
构建能共同表达日本血吸虫 2 3kDa膜抗原 (Sj2 3)与细胞因子IL 12的多价DNA疫苗 .RT PCR的方法从日本血吸虫成虫获得Sj2 3的基因片段 ,克隆到载体 pVIVO2 mIL12 /mcs上 ,进行酶切和测序鉴定 .采用免疫荧光的方法检测多价DNA疫苗 pVIVO2 Sj2 3 IL12在小鼠骨骼肌细胞的表达 .成功地构建了能共同表达日本血吸虫 2 3kDa膜抗原 (Sj2 3)与细胞因子IL 12的多价DNA疫苗 .此多价DNA疫苗在免疫小鼠肌细胞的细胞膜和细胞浆均获得了Sj2
To construct multivalent DNA vaccine coexpressing Sj2 3 and cytokine IL-12, DNA fragment encoding the Sj23 gene of schistosoma japonicu m was obtained from the adult worm by RT-PCR. The gene was cloned into expresse d plasmid pVIVO2-mIL12/mcs, and identified by restriction analysis and sequence . The expression of Sj23 in muscle tissue was determined by Immunofluorescence a ssay. Multivalent DNA vaccine which co-expresses Sj23 and cytokine IL-12 was s uccessfully constructed. Sj23 was expressed on the membrane and in the plasma of muscle cells of the mice immunized with pVIVO2-Sj23-IL-12.
