鼻咽良恶性病变中EBV-LMP1、CyclinB1、P34^(cdc2)和Survivin的表达及意义

Expression of EBV-LMP1,cyclinB1,P34^(cdc2) and survivin in patients with nasopharyngeal diseases

目的 :探讨P34cdc2 、CyclinB1和Survivin在鼻咽粘膜慢性炎、鼻咽癌和颈部淋巴结转移灶中的表达及意义 ,并分析与EBV LMP1表达的相互关系。方法 :应用免疫组化检测 30例鼻咽粘膜慢性炎、30例鼻咽癌和 30例鼻咽癌颈部淋巴结转移灶中LMP1、P34cdc2 、CyclinB1和Survivin的表达。结果 :LMP1、P34cdc2 、CyclinB1和Survivin在鼻咽粘膜慢性炎中的阳性率分别为 2 0 .0 % (6 /30 )、10 .0 % (3/30 )、10 .0 % (3/30 )和 3.3% (1/30 ) ;在鼻咽癌中的阳性率分别为 6 3.3% (19/30 )、73.3% (2 2 /30 )、5 0 .0 % (15 /30 )和 36 .7(11/30 ) ,与鼻咽粘膜慢性炎比较差异均有显著性 (P <0 .0 1) ;在鼻咽癌颈部淋巴结转移灶中的阳性率分别为 70 0 % (2 1/30 )、80 .0 % (2 4 /30 )、76 .7% (2 3/30 )和 4 3.3% (13/30 ) ,与鼻咽粘膜慢性炎比较差异均有显著性 (P <0 .0 1) ,与鼻咽癌相比较均有所增高 ,但仅CyclinB1有统计学意义(P <0 .0 5 )。在鼻咽癌及其颈部淋巴结转移灶中 ,LMP1与P34cdc2 和CyclinB1的表达均有显著相关性 (分别P <0 .0 1,P <0 .0 5 )。结论 :鼻咽癌中存在P34cdc2 、CyclinB1和Survivin蛋白过表达 ,LMP1与P34cdc2 和CyclinB1可能协同致病。

Objective:To investigate the expressions of P34 cdc2,cyclin B1 and survivin in the lesions of chronic nasopharyngitis,nasopharyngeal carcinoma(NPC)and nodal metastasis of NPC,and their relation to EBV-LMP1 expression.Methods:The expressions of EBV-LMP1,P34 cdc2,cyclin B1 and survivin were detected by immunohistochemical method in the paraffin-embedded specimens of 30 chronic nasopharyngitis,30 NPCs and 30 lymph-node metastases.Results:The expressions of EBV-LMP1,P34 cdc2,cyclin B1,and survivin in NPC and nodal metastasis were superior to those in chronic nasopharyngitis(P<0.01).Only cyclin B1 expression was up-regulated in nodal metastasis in comparison with NPC (P<0.05).EBV-LMP1 expression in NPC and nodal metastasis was significantly correlated with expressions of P34 cdc2 (P<0.01) and cyclin B1 (P<0.05),respectively.Conclusion:EBV-LMP1,P34 cdc2,cyclin B1 and survivin are all overexpressed in NPC.EBV-LMP1 may be cooperated with P34 cdc2 and cyclin B1 in the development of NPC.
