改善血管桥舒张功能的实验研究:常温下尼可地尔对大隐静脉血管桥内皮源性超极化因子介导血管舒张功能的保护作用 被引量:2

摘要 目的:探讨常温下尼可地尔(nicorandil,NCR)和罂粟碱对大隐静脉血管桥血管内皮超极化因子(endothelium-derivedhyperpolarizingfactor,ED-HF)介导血管舒张功能的影响。方法:采用器官槽法研究在37℃有氧条件下用重碳酸盐缓冲液(KH)、含0.1mmol/LNCR的KH、含1.2mmol/L罂粟碱的KH及在0.1mmol/LNCR的KH中分别加入0.12,1.2,12mmol/L罂粟碱浸泡血管环1h后,再用7μmol/L吲哚美辛(indomethacin,Indo)和300μmol/LN-硝基-L-精氨酸作用后,检测30nmol/LU46619及不同浓度A23187引发的血管收缩舒张反应。结果:单纯浸泡于KH中的血管环,A23187引发的血管舒张反应为(66.54±2.40)%,在含1.2mmol/L罂粟碱的KH和含12mmol/L罂粟碱加NCR的KH中,A23187引发的血管舒张反应分别为(31.36±2.27)%和(37.71±6.59)%,与前者相比,后两者明显减低;在含0.1mmol/LNCR的KH和含1.2mmol/L罂粟碱加NCR的KH中,A23187引发的血管舒张反应分别为(64.39±5.69)%和(63.09±6.51)%,与单纯浸泡于KH中的血管环无明显差异。电镜所见罂粟碱溶液损害血管内皮细胞,随浓度加深,损害程度加重。含NCR的罂粟碱KH内皮细胞损害减轻。结论:加入了NCR的1.2mmol/L罂粟碱溶液能很好的舒张血管环,并对EDHF介导血管舒张功能有保护作用,高浓度罂粟碱溶液破坏血管内皮,并? AIM:To explore the protective effect of nicorandil(NCR) and papaverine on saphenous vein graft(SVG) endothelium derived hyperpolarizing factor (EDHF) mediated vasodilatation under normal temperature. METHODS: The organ chamber technique was used.The vascular rings were incubated at 37 ℃ for 1 hour in the following different aerobic Kerbs Herseleit solution(KH):KH only,KH with 0.1 mmol/L NCR,KH with 1.2 mmol/L papaverine,KH with 0.1 mmol/L NCR and 0.12,1.2 and 12 mmol/L papaverine respectively.Then 7 μ mol/L indomethacin(Indo) and 300 μ mol/L NG nitro L arginine(LNNA) were added to the above KHs.The vasoconstriction and vasodilatation responses to 30 nmol/L U46619 and A23187 of different concentration were then assessed. RESULTS: The ring responses to A23187 were(66.54± 2.40)% in KH only,(31.36± 2.27)% in KH with 1.2 mmol/L papaverine and( 37.71± 6.59) % in KH with 0.1 mmol/L NCR and 12 mmol/L papaverine,compared with that in KH only,the vasodilatation declined significantly in the latter two.Compared with that in KH only,no significant difference was observed in KH with 0.1 mmol/L NCR[(64.39± 5.69) % ]and KH with NCR and 1.2 mmol/L papaverine[(63.09± 6.51) % ].Under electron microscope, endothelial cell damage caused by papaverine depended on the concentration of papaverine,and the damage reduced in presence of NCR. CONCLUSION: Saphenous vein rings can be relaxed in KH with NCR and 1.2 mmol/L papaverine,which has the protective effect on EDHF mediated vasodilatation.High concentration of papaverine has the deleterious effect on vascular endothelium and inhibition to vasodilatation mediated by EDHF.
