摘要
目的 分别以 CD1 0 5、CD34作为血管生成的标记物 ,对照研究大肠腺癌组织中血管生成与肿瘤细胞凋亡的关系。方法 应用免疫组化 S- P法 ,对 6 2例大肠腺癌组织中血管内皮细胞的 CD1 0 5、CD34抗原进行染色 ,检测肿瘤内微血管密度 ;对肿瘤细胞增殖核抗原 (PCNA)染色检测增殖指数。应用 TU NEL法检测凋亡细胞 ,并计算凋亡指数。结果 以 CD34作为血管生成的标记物 ,平均凋亡指数在有较低微血管密度的患者和有较高微血管密度的患者间差异无显著性 (分别为 4 .6 7± 2 .0 7和 4 .1 6± 1 .85 ,P=0 .31 5 )。相反 ,以 CD1 0 5作标记物 ,平均凋亡指数在较低微血管密度的患者显著高于有较高微血管密度的患者 (分别为 5 .0 3± 2 .1 0和 3.85± 1 .6 7,P =0 .0 1 7)。 CD34或CD1 0 5与肿瘤细胞增殖活性无显著相关性。
Objective To study the correlation between tumor cell apoptosis index(AI)and intratumoral microvessel density(IMVD)in colorected adenocarcinoma by CD105 and CD34 as two different markers of angiogenesis.Methods Sixty two surgical specimens from colorectal adenocarcinoma tissue were studied.IMVDs were determind by staining the antigens of CD34 and CD105 immunohistochemically(S P method).Apoptosis cells were estimated by TUNEL method and AI calculated.Results When CD34 was used as a marker of angiogenesis,the mean A in the lower IMVD and the higher IMVD patients were 4.67±2.07 and 4.16±1.85 respectivly,which demonstrated no significant deffernce between the two group of patients.In contrast,when CD105 was used,the mean AI in the lower IMVD patients was higher than that in the higher IMVD patients significantly.(5.03±2.10 and 3.85±1.67 respectivly; P =0.017<0.05).There was no significant correlation between proliferative activity and CD34 IMVD or CD105 IMVD.Conclusion These results illustrated that the decreased angiogenesis may induce enhancement of apoptosis of the tumor cell without influence of cell proliferation.
