摘要
目的 研究呱氨托美汀对小鼠胃黏膜的保护作用。方法 以托美汀 (TOL)、呱氨托美汀 (AMG)高中低三个剂量小鼠连续灌胃给药 7d ,以羧甲基纤维素钠为对照组 ,观察小鼠胃损伤情况及体重增长情况。以CMC、TOL、AMG高中低三个剂量组等容灌胃 (0 5mlig) ,1h后给 5 0 %乙醇ig0 5ml/只 ,造成乙醇性胃黏膜损伤模型。 1h后分离小鼠胃并以福尔马林固定 ,观察胃损伤情况。结果 AMG多剂量给药及单剂量灌胃后乙醇诱导的胃损伤模型中胃损伤评分结果与溶剂对照组比较差异无显著性 ,与托美汀组相比具有明显的胃保护作用。且连续灌胃 7d ,小鼠体重增长情况AMG组及CMC对照组给药后较给药前明显增长 ,TOL组差异无显著性。
Objective To investigate gastroprotective effect of amtolmetin guacyl in mice. Methods Six groups of ten mice each received CMC, TOL or AMG(75, 150, 300 mg/kg) daily for 7 days. Six hours after the last administration,the mice were killed to observe the severity of the gastric mucosal damage and the variation of the weights. Another six groups of eight mice each were given NS, CMC, TOL or AMG (75, 150, 300 mg/kg) pretreated,1 hour later the mice received 50% ethanol(0 5 ml/mouse intragastrically). And 1 hour later the mice were killed. The stomachs were removed and checked. Results AMG demonstrated significant gastroprotective effect in the former two tests after AMG(300, 150, 75 mg/kg) as the were intragastrically administered in the mice. Moreover the intensity of gastroprotective effect increased in an dose-dependent manner. TOL did not manifest the same effect as AMG. After being intragastrically administered with AMG and CMC for 7 days, the murine weights were not decreased, while the mouse intragastrically administered with IND weighted significantly less. Conclusion These data indicate that amtolmetin guacyl has significant gastroprotective effect.
