摘要 目的 探讨环孢素A治疗急性排斥反应的作用机制。方法 采用Ono方法建立SD大鼠心脏移植排斥反应动物模型 ,将实验分为两组 :对照组n =10 ,未进行任何治疗 ;环孢霉素A(CsA)组 :n =2 0 ,术后 1~ 7d腹腔内注射CsA 10mg/ (kg·d)。采用ELISA法测定术后IL 2的含量 ,采用Tunel法检测心肌细胞凋亡 ,并计算凋亡指数。结果 两组移植物存活时间为 7 2 5d± 0 71d和 19 4d± 2 0 7d ,实验组比对照组延长 12d(P <0 0 1)。实验组术后IL 2含量明显减少 ,与对照组相比较差异有显著性 (P <0 0 5 )。两组中心肌细胞凋亡指数分别为 2 84± 1 4 8和 0 5 4± 0 19(P <0 0 0 0 1)。结论 CsA通过抑制IL 2的分泌和心肌细胞凋亡 ,发挥抗排斥作用。 Objective To study the effect of Cyclosporin A(CsA) on acute rejection in rat's heart transplantation. Methods Heart transplant rejection model in rats was set up by Ono's method. The experimental objects were divided into two groups. Control group: n =10 without any treatment, experimental group: n =20 intraperitoneal injection of CsA at dose of 10 mg/(kg/d ) 1~7 d after operation. Concentration of IL-2 after operation was tested by ELISA. Cell apoptosis was measured by Tunel method. Apoptosis index (AI) was calculated. Results The survival time of allograft in the two groups were (7 25±0 71) and (19 4±2 07) d respectively. CsA prolonged 12 days of survival time in the experiment group than the control group( t =9 393, P <0 0001). The concentration of IL-2 in CsA group was more significantly decreased after operation than that in the control group ( F =20 42 P <0 05). Myocyte apoptosis index in the two groups were 2 84±1 48 and 0 54±0 19 respectively. There was significant difference in the two groups (t=5 745 P <0 0001). Conclusion CsA can inhibit secretion of IL-2 and myocyte apoptosis and have anti-rejection effect.
