摘要
目的 给予雌性去势大鼠雌激素替代治疗后观察血清雌二醇水平和脑组织抗氧化能力之间的关系,从而探讨雌激素对中枢神经系统的作用及可能机制。方法 清洁级9月龄雌性SD大鼠30只随机平均分为①用药组(去势后每日给予共轭雌激素0.2 mg/kg/d灌胃);②对照组;③假手术组。12周后测定血清E_2水平、大脑额叶皮质及海马组织中超氧化物歧化酶(SOD)、丙二醛(MDA)、总抗氧化能力(T-AOC)、活性氧(ROS)水平等生化指标。采用非配对t检验对以上指标进行统计学分析。结果 1.用药组血清E_2水平显著高于对照组及假手术组(均P<0.001);假手术组E_2水平高于对照组但差别无显著性(P>0.05)。2.用药组及假手术组大脑额叶皮质SOD活力均高于对照组(均P<0.05),用药组与假手术组之间差异无显著性(P>0.05);用药组海马组织中SOD活力高于对照组(P<0.05),用药组与假手术组及对照组与假手术组之间差异无显著性(P>0.05)。3用药组大脑额叶皮质MDA水平显著低于对照组及假手术组(P<0.05),对照组及假手术组之间差异无显著性(P>0.05);用药组海马组织中MDA水平也显著低于对照组(P<0.005)而与假手术组之间及对照组与假手术组之间差异均无显著性(P>0.05)。4.用药组及假手术组大脑额叶皮质及海马组织中总抗氧化能力均显著高于对照组(P<0.05),用药组及?
Purpose: To discuss the effect on central neural system and the probable mechanism of estrogen through observing the relationship between serum E 2 level and brain antioxidation capacity of emasculated female rats after HRT. Methods: Thirty female SD rats were divided randomly and averagely into 3 groups: experimental group (underwent oophorectomy and then received conjugated estrogen 0.2 mg/kg/d), control group and SHAM group. After 12 weeks, serum E2, brain NO, NOS, SOD, MDA, T-AOC and ROS were determined, and the relationship of them was analysed. Results: 1 Experimental group had a higher serum E2 level than control group and SHAM group. 2 Experimental group and SHAM group had higher levels of brain SOD and T-AOC than control group. 3 Experimantal group had a lower level of brain MDA than control group and SHAM group. 4 Experimental group and SHAM group had lower levels of brain ROS than control group. Conclusions: HRT augments brain antioxidation capacity of emasculated female rats, so as to give protection to neurons.
