摘要
目的 :探讨梗死脑组织上清对骨髓基质细胞分化的诱导作用。方法 :从正常人肋骨分离培养骨髓基质细胞 ,以梗死脑组织上清诱导骨髓基质细胞 ,相差显微镜以及免疫荧光染色检测诱导结果。结果 :诱导后骨髓基质细胞出现分化 ,胞体呈锥形 ,细胞有长突起 ,交织成网。免疫荧光染色显示诱导后的骨髓基质细胞表达MAP 2和GFAP的比例明显高于其他组 (P <0 .0 5 ) ,表达MAP 2的细胞明显多于表达GFAP阳性细胞(P <0 .0 5 )。结论 :梗死脑组织上清可以诱导骨髓基质细胞向神经细胞分化。
Objective:To study the effect of infarcted cerebral constitution on the differentiation of human bone marrow stromal cells into neural cell.Methods:hMSCs from healthy person were propagated in DMEM with 10%FBS. Infarcted cerebral constitution get from middle cerebral artery occlusion rat was used to induce hMSCs. Phase contrast microscope were applied to detect the differentiation.Results:After induction,cells expressed pyramidal cells bodies and elongated processes. MAP-2 positive cells and GFAP positive cells are much more in experimental group than others(P<0.05).MAP-2 positive cells are much more than GFAP positive cells(P<0.05).Conclusion:Infarcted cerebral constitution can induce human bone marrow stromal cells into neural cell.
