苯酚-硫酸光度法测定细脚拟青霉多糖的含量

Determination of polysaccharides in Paecilomyces tenuipes with spectrophotometry of phenylhydrate-sulfuric acid

目的 :测定细脚拟青霉中多糖的含量。方法 :应用苯酚 硫酸光度法测定细脚拟青霉总多糖制品中的糖含量。结果 :测定波长为 4 90nm ,在 2 0～ 2 0 0mg·L-1范围内糖含量与吸光度有良好的线性关系 ,回归方程为 :A =0 .0 0 75 72C +0 .0 6 0 12 ,r=0 .9994。平均回收率为 (97.9± 1.6 9) % ,RSD =2 .70 %。测得细脚拟青霉总多糖制品中糖含量为 76 .10 % (n =5 )。结论 :该法操作简便、可靠 ,样品显色稳定 ,重现性好 ,可用于细脚拟青霉多糖含量的测定。

Objective:To detect the polysaccharides in Paecilomyces tenuipes.Methods:The polysaccharide content was detected with phenylhydrate-sulfuric acid method.Results:The detectde wavelength was 490 nm.Linear range was 20～200 mg·L -1(A=0.007572C+0.06012,r=0.9994). The average recovery rate was 97.9±1.69% and RSD was 2.70%. The polysaccharide content of Paecilomyces tenuipes in the total polysaccharides was 76.10%(n=5).Conclusion:The method was convenient and reliable,and with satisfying stability and repeatability to be used.
