摘要 目的 探讨腺病毒E1A基因对吸烟所致肺泡上皮细胞IL-8表达的影响。方法 通过脂质体转染的方法,将腺病毒E1A基因转染肺泡上皮细胞(A549细胞),经G418筛选、Western blot鉴定E1A阳性表达A549细胞单克隆,不同浓度的香烟提取物(CSE)活化,测定细胞IL-8 mRNA表达和上清液中IL-8浓度。结果 与未转染和对照质粒转染A549细胞相比较,经CSE活化后E1A阳性表达A549细胞IL-8 mRNA表达和蛋白释放显著增加。结论 腺病毒E1A基因显著增加吸烟所致的肺泡上皮细胞IL-8表达,提示腺病毒潜伏感染可能通过放大吸烟所致的气道炎症反应在慢性阻塞性肺疾病的发病中发挥重要作用。 Objective To explore the influence of adenovirus El A gene on IL-8 expression induced by cigarette smoke extract (CSE) in alveolar epithelial cells. Methods To transfect human alveolar epithelial cells ( A549 cells) with plasmids carrying the adenovirus El A gene and isolate stable transfectants expressing El A protein by Western blot. A549 cells, A549 cells transfected with control plasmids and ElA-producing clones were tested the IL-8 expression after CSE stimulation. Results As compared with parental cells or cells transfected with control plasmids, IL-8 mRNA expression and protein release were markedly increased by CSE stimulation in ElA-positive A549 cells. Conclusion Adenovirus El A gene can markedly increase IL-8 expression induced by CSE in the alveolar epithelial cells. This suggests it may play an important role in the pathogenesis of COPD through amplifying the inflammatory process present in the airway of the smokers.
