摘要 目的 体外实验探讨人类巨细胞病毒(HCMV)与细胞表面硫酸乙酰肝素蛋白聚糖(HSPGs)结合后对人类白细胞抗原I(HLA-I)表达的影响。方法 流式细胞仪检测不同干预条件细胞表面HLA-I的表达。免疫组织化学法检测不同干预条件细胞内HCMV pp65蛋白的表达。结果 流式细胞仪检测的结果表明人胚肺成纤维细胞(HELF)表面HLA-I的表达HCMV感染组低于对照组;紫外线灭活HCMV(UV-HCMV)组高于对照组;肝素和氯酸钠干预HELF细胞后再用UV-HCMV感染的HELF细胞,其表面HLA-I的表达低于UV-HCMV组。免疫组织化学法检测结果显示:HCMV感染HELF 24 h,HCMV pp65蛋白呈阳性表达;UV-HCMV感染HELF 24 h,HC-MV pp65蛋白呈阴性表达。结论 HSPGs可能参与影响HLA-I在HCMV感染的HELF细胞表面上的表达,对HCMV逃避宿主的免疫监视起间接调节作用。 Objective To investigate the effect of human cytomegalovirus (HCMV) binding to heparan sulfate proteoglycans on the expression of human leucocyte antigen class I (HLA- I ) on the surface of human embryo lung fibroblasts (HELF) in vitro. Methods Flow cytometry was carried out to assay the expression of HLA- I on the surface of HELF prepared by different methods. Inmmunocytochemistry was performed to examine the expression of HCMV pp65 protein in HELF infected by different methods. Results The expression of HLA- I on the surface of HCMV infected HELF group was less than that of the control group. The expression of HLA- I on the surface of UV- HCMV infected HELF group was strong than that of the control group. After HELF prepared by heparin or sodium chlorate was infected by UV-HCMV, the expression of HLA-I on the surface of UV-HCMV infected HELF group was decreased. HCMV pp65 protein was positive after HELF had been infected by HCMV for 24 h. HCMV pp65 protein was negative after HELF had been infected by UV-HCMV for 24 h. Conclusion HSPGs probably influence the expression of HLA- I on the surface of HCMV infected HELF. HSPGs indirectly regulate the HCMV immune surveillance evasion.
