摘要 目的 研究甲基化抑制剂5-氮-2’-脱氧胞苷对胆管癌细胞株生长周期及凋亡的影响,初步探讨其应用于临床治疗的可能性。方法 应用MTT法检测不同浓度的5-氮-2’-脱氧胞苷对胆管癌细胞株QBC939存活率的影响,应用流式细胞术检测5-氮-2’-脱氧胞苷对QBC939细胞生长周期及凋亡率的影响。结果 5-氮-2’-脱氧胞苷在浓度为0.5μmol/L时即可以抑制胆管癌细胞QBC939的增殖,24 h半数致死量为5.0 μmol/L,细胞周期中处于G0/G1期的细胞比例增多,凋亡的发生率增高,而且以上作用与药物浓度及作用时间在一定范围内呈正相关。结论 5-氮-2’-脱氧胞苷可能通过消除某些抑癌基因的启动子甲基化状态,使其重新表达而抑制胆管癌细胞的生长,并促进其凋亡。 Objective To study the effects of methylation inhibitor 5-aza-2 -deoxycytidine on the growth and apoptosis of bile duct cancer cell line QBC939, and the possibility of its application in clinical treatment. Methods The survival rate and apoptosis rate of QBC939 after being treated with different density of 5-aza-2'-deoxycytidine were detected by using the method of MTT and flow cytometry. Results 5-aza-2'-deoxycytidine at a concentration of 0. 05 mmol/L could inhibit the proliferation of bile duct cancer cell line QBC939 and the LC50 was 5. 0 mmol/L within 24 h. The number of cells in the G0/G1 was increased and the apoptotic rate was also increased. The above effects of 5-aza-2'-deoxycytidine were concentration and time dependent in a range. Conclusion 5-aza-2'-deoxycytidine can inhibit the growth of QBC939 and enhance the apoptosis by eliminating the promotor methylation state of some tumor inhibitory genes, especially some cell cycle control genea.
