摘要
目的 建立新的稳定 5 0 %大鼠部分肝移植的模型上 ,研究不同冷缺血时间对移植术后存活率和肝再生的影响。方法 A组为对照组 :5 0 %肝切除 ;实验组 :5 0 %部分肝移植组 (B、C、D组冷缺血时间分别为 4 5 min、4 h和 1 0 h)。观察各实验组生存率 ,并分别于移植术后 2 4 h、4 8h、96 h免疫组化检测各实验组增殖细胞核抗原 (PCNA)的表达 ,采用统计学的方法分析不同冷缺血时间对大鼠部分肝移植术后 2 4小时存活率和肝再生的影响。结果 B、C、D组存活率分别为 80 %、4 5 %、2 5 % ;PC-NA表达在术后 4 8小时达到峰值分别为 5 4± 5 .2 ;4 4± 3.0 ;2 0± 1 .9。D组 PCNA表达明显降低 (P<0 .0 5 )。结论 部分肝移植术后肝再生峰值较肝切除后延迟 。
Objective To evaluate survival rate of different cold preservation time in a novel model of partial liver transplantation in rats.Methods Wister rats subjected to 50% liver resection (control:group A) were compared with rats 50% partial liver transplantation with different cold ischemia.Livers were preserved at 4 degrees C for 45 minutes(group B),4 hours (group C)and 10 hours (group D) in University of Wisconsin solution.The expression of the proliferative cell nuclear antigen (PCNA) of liver in rats were evaluated by immune histochemical method 1d,2d,4d after operation,repectively.Result The survival rate with group B,group C.group D was 80%,45%,25%,and the expression of PCNA peak was 54±5.2;44±3.0;20±1.9 at 48h after transplantation,respectively.Prolonged time of cold preservation(10 hours)was associated with a dramatic decrease of PCNA ( P <0.05).Conclusion The expression of PCNA peak in partial liver transplantation slightly delayed compared with hepatectomy.Prologed cold preservation significantly decreased the regenerative ability of the liver and animal survival in partial liver transplantation in the rats.
