摘要
目的 以磷酸钙骨水泥(CPC)为载体,制备阿霉素—磷酸钙骨水泥药物缓释体系,观察其体外药物释放情况。方法 将阿霉素(ADM)包埋在磷酸钙骨水泥中,固化后的骨水泥进行体外药物溶出试验,用荧光光谱法研究药物在37℃模拟体液中的释放情况。结果 ADM可从载药磷酸钙骨水泥中缓慢释放,周期在1个月以上,药物的释放速度和浓度与CPC的药物包埋量有关,药物释放基本符合Higuchi等所建立的模型。结论 以CPC为载体,对ADM进行适当的包埋,可制备稳定的药物缓释体系,阿霉素—磷酸钙骨水泥缓释体系具备有修复骨缺损和药物治疗的双重功能。
Objective To develop a novel drug delivery system hased on calcium phosphate cement (CPC) with
adrimycin (ADM) as a model drug and to study its drug release test in vitro. Method ADM was loaded on CPC, after
harden,fluorescance spectrum was used to measure in vitro drug release from CPC pellets into simulated body fluid at
37℃.Result ADM could be released slowly and stably from the ADM - loaded CPC above 1 month, the drug release
rate and concentration of ADM - loaded CPC is concerned with the quantity of ADM, the drug release was found to agree
with the higuchi equation. Conclusion A stable drug delivery system could be developed by loading ADM into CPC and
the controlled delivery of drug could be attained by choosing the reasonable implanting operation and adjusting the param-
eters such as the amount of drug, this system has the effect on both bone repair and medicational therapy.
