上下呼吸道炎症反应相关性研究 被引量:39

摘要 目的 通过变应性鼻炎和变应性哮喘动物模型探讨上下呼吸道炎症反应的相关性及其机制。方法 采用卵清蛋白致敏并激发制成变应性鼻炎和变应性哮喘模型。实验动物肺功能检测仪检测肺功能、HE染色和甲苯胺蓝染色分别检测鼻黏膜和肺组织中嗜酸粒细胞和肥大细胞 ,免疫组化检测上述组织中的血管细胞黏附分子 Ⅰ (vascularcellularadhesionmolecular Ⅰ ,VCAM Ⅰ )和白细胞介素 13 (interleukin 13 ,IL 13 )的表达。以酶联免疫吸附实验 (enzyme linkedimmunosorbentassay ,ELISA)检测外周血IL 5浓度。结果 变应性鼻炎和变应性哮喘模型鼻黏膜和肺组织中嗜酸粒细胞、肥大细胞明显多于对照组 ,变应性鼻炎组和变应性哮喘组肺功能 0 3s率显著低于对照组 ,前两组肺组织中VCAM Ⅰ和IL 13阳性血管数显著多于对照组。变应性鼻炎组和变应性哮喘组外周血IL 5浓度显著高于对照组 ,并且与嗜酸粒细胞的数量呈显著正相关。结论 上下呼吸道炎症反应具有一致性 ,变应原反复激发上呼吸道也能引起下呼吸道炎症改变和功能减退。 Objective To evaluate the correlation between upper and lower respiratory inflammation Methods Thirty rats were randomly divideded into allergic rhinitis group (AR), asthma group (AS), and control group(Con) The pulmonary function was measured by pulmonary function instrument of animal model, the infiltration of eosinophils and mast cells was detected by hematotoxylin-eosin staining (HE staining) and toluidine blue staining respectively, the expression of VCAM-Ⅰ and IL-1 3 was examined by immunohistochemistry ELISA was used to measure the concentrat ion of IL-5 in the peripheral serum Results The numbers of eosinophils and mast cells in nasal mucosa and lung tissue of AR were both significantly higher than those of Con, and similar results were observed between AS and Con The forced expiratory volume in 0 3 second (FEV0 3)/ forced vital capacity (FVC) of AR and AS was both significantly lower than that of Con The numbers of VCAM-Ⅰ and IL-13 positive vessels in lung tissue of AR were both significantly higher than those of Con, and similar results were observed between AS and Con The concentration of IL-5 in the serum of AR and AS was both significantly higher than that of Con The concentration of IL-5 in serum was positively correlated with the number of eosinophils in peripheral blood of AR and AS Conclusion The inflammation is similar between AR and AS, and repeatedly challenge upper re spiratory tract can impact on lower respiratory response
