摘要
目的探讨p16蛋白在胶质细胞瘤发生发展以及肿瘤复发中的作用。方法采用免疫组织化学LsABC法检测p16蛋白在正常脑组织、瘤旁脑组织和不同病理级别胶质细胞瘤组织中的表达,并用图像分析系统定量测定p16蛋白的表达面积;统计分析肿瘤复发前后p16蛋白表达的变化及其与复发时间的相互关系。结果(1)正常脑组织、瘤旁脑组织和胶质细胞瘤组织中p16蛋白的表达面积依次下降,分别为(613.43±217.21)μm2、(560.26±202.89)μm2和(307.24±116.91)μm2(P<0.05或P<0.01);(2)随着胶质细胞瘤病理级别的升高,p16蛋白的表达面积亦依次下降,星形细胞瘤(Ⅰ~Ⅱ级)为(491.91±215.32)μm2,间变性星形细胞瘤(Ⅲ级)为(396.83±201.87)μm2,原发多形性胶质母细胞瘤(Ⅳ级)为(203.67±143.19)μm2(P<0.05或P<0.01);(3)肿瘤复发者p16蛋白的表达面积减小,而复发后病理级别升高者p16蛋白表达面积差值犤(103.152±20.829)μm2犦明显高于未升高者犤(59.381±16.215)μm2犦,差异具有显著性意义(P<0.01);(4)肿瘤复发间隔时间与p16蛋白的表达水平呈负相关(r=?0.453,P<0.01)。结论p16蛋白的表达与胶质细胞瘤恶性进程和复发有关,进一步研究p16蛋白表达水平的改变机制,将有助于对胶质细胞瘤复发的防治。
Objective To study the role of p16protein in glioma development,progression and recur-rence.Methods The p16protein expression was determined in normal cerebral tissue,peritumoral tissue and different grade gliomas by LsABC(labelled streptavidin biotin method)immunohistochemical method,and its distribution areas were measured by image analyzing system;the expression alternations in pre-and post-re-currence of tumors and the correlation with recurrent time were statistically analyzed.Results1)The ex-pression of p16protein decreased in the order of normal cerebral tissue,peritumoral tissue and glioma,the distribution areas were(613.43±217.21)μm 2 ,(560.26±202.89)μm 2 and(307.24±116.91)μm 2 respectively(P<0.05or P<0.01);2)The p16protein expression decreased as the pathological grade increased,the distribution areas of astrocytoma (gradeⅠ-Ⅱ),anaplastic astrocytoma (gradeⅢ),and polymorphic primary glioblastoma (gradeⅣ)were(491.91±215.32)μm 2 ,(396.83±201.87)μm 2 and(203.67±143.19)μm 2 respectively(P<0.05or P<0.01);3)The distribution areas of p16protein expression decreased in recurrent tumors,especially the p16protein expression area difference was significant higher in those with increased grade(103.152±20.829)μm 2than that in unchanged grade(59.381±16.215)μm 2 (P<0.01);4)The recurrent interval time was negatively correlated with p16protein expression level(r=?0.453,P<0.01).Conclusion The p16protein expression is correlated with progression and recurrence of gliomas,thus further study on mechanism of its expression alter-nation will be helpful to the prevention and treatment of glioma recurrence.
