大鼠动脉粥样硬化时血小板内一氧化氮合酶的表达及其活性 被引量:2

摘要 目的 :研究动脉粥样硬化形成时血小板内一氧化氮合酶 (NOS)的表达特点和活性。 方法 :大鼠分为 3组 :第 1组为正常对照组 ,以普通饮食饲养 ;第 2组饲以高脂饲料 (不含维生素 D3) ;第 3组为维生素 D33× 1 0 5U/ kg一次肌内注射、球囊损伤主动脉内皮和饲以含维生素 D31 .2 5× 1 0 6 U/ kg的高脂饲料。 90 d后检测大鼠血小板内 NOS的表达和活性。结果 :第 2组和第 3组大鼠血小板中内皮型 NOS(e NOS)均较第 1组表达减弱、活性下降 (P<0 .0 1 ) ,而第 2、3组之间无统计学差异 ;诱导型 NOS(i NOS)表达和活性在第 2组与第 1组间无统计学差异 ,但第 3组较前两组均显著增加 (P<0 .0 1 )。结论 :结果提示 ,动脉粥样硬化时血小板 e NOS表达和活性受到抑制 ,而 i Objective:To investigate the expression and activity of nitric oxide synthase in platelets in the rats with atherosclerosis. Methods: Rats were divided into 3 groups: rats in group 1 were fed with normal diet,group 2 were fed with high cholesterol diet(no vitamin D 3), group 3 were treated with intramuscular injection of vitamin D 3 (3×10 5 U/kg) and the endothelium of the arteries were denudated,followed by high cholesterol diet.The expression and activities of nitric oxide synthase in platelets in the 3 groups were investigated 90 d later. Results: Compared with group 1,group 2 and group 3 had less expression and lower activities of endothelial nitric oxide synthase( P <0.01); there were no differences between the 2 experimental groups. For inducible nitric oxide synthase,there were no differences between group 1 and group 2,whereas the expression and activities in group 3 were higher than those in the first 2 groups( P <0.01). Conclusion: There are endothelial and inducible nitric oxide synthase in platelets. Expression and activity of endothelial nitric oxide synthase decrease in atherosclerosis,and those of inducible nitric oxide synthase are correlated with the development of atherosclerosis.
