摘要 目的 :研究缺血、血管活性物质对肾脏 Na+ ,K+ - ATP酶活性的作用。方法 :选用 SD大鼠 ,用左肾动脉灌注法制备肾缺血模型。按左肾动脉处理性质不同分以下 7组 :(1 )正常组 (n=7) ;(2 ) 6 0 m in钳夹组 (n=5 ) ;(3)去甲肾上腺素 (NE)灌注组 (n=5 ) ;(4 ) NE加酚妥拉明 (PH)灌注组 (n=5 ) ;(5 )内皮素 (ET)灌注组 (n=7) ;(6 ) ET+PH灌注组 (n=7) ;(7)吲哚美辛 (IND)静脉注射加 ET灌注组 (n=7)。术中监测动物血压 ,术后迅速取肾组织制备匀浆 ,测定 Na+ ,K+ - ATP酶活性 (μmol· mg- 1 · h- 1 )。 结果 :实验中系统血压无大变化。正常组动物左右肾 Na+ ,K+ - ATP酶活性无差别 (4 .1 8± 1 .4 1 vs3.6 3± 1 .2 2 ,P>0 .0 5 ) ;钳夹左肾动脉6 0 min,其左肾酶活性较对侧明显下降 (1 .96± 0 .4 2 vs5 .33± 0 .77,P<0 .0 0 1 ) ;左肾动脉灌注 NE,左肾酶活力明显下降 (2 .5 9±0 .4 9vs3.73± 0 .81 ,P<0 .0 5 ) ;而 NE与 PH同时左肾动脉灌注时 ,酶活性无改变 (3.80± 0 .4 0 vs3.70± 0 .5 2 ,P>0 .0 5 ) ;左肾灌注 ET处理侧酶活性显著降低 (2 .35± 1 .0 5 vs3.6 0± 0 .99,P<0 .0 5 ) ,ET与 PH同时灌注其结果同 ET组 ,较处理侧酶活性也显著降低 (2 .35± 0 .88vs3.4± 0 .85 ,P<0 .0 1 ) ,ET左肾动脉灌注? Objective:To study the effect of ischemia and vasoactive substances on renal Na +,K + ATPase activities. Methods: The right kidney of SD rat was used as control and the left as experimental group.Left renal arteries were clamped for 60 min to induce ischemia or subjected to different vasoactive substances. The rats were divided into following groups: control group; renal artery clamp(RAC) group: left renal arteries were clamped for 60 min; norepinephrine (NE) group; NE + phentolamine (PH) group; endothelin (ET) group; ET+PH group; indomethacin(IND) +ET group. The activities of Na +,K + ATPase(μmol·mg -1 ·h -1 ) were measured in renal preparations.Results: The system blood pressure was smooth throughout the experiment.There were no significant differences in the Na +,K + ATPase activities between the 2 kidneys in control group (4.18±1.41 vs 3.63±1.22, P >0.05), NE+PH group (3.80±0.40 vs 3.70±0.52, P >0.05) and IND+ET group (2.7±0.89 vs 2.83±0.74, P >0.05). In RAC group, the Na +,K + ATPase activity of the left kidney decreased compared with that of the right kidney (1.96±0.42 vs 5.33±0.77, P <0.001), and so were the Na +,K + ATPase activities in NE group (2.59±0.49 vs 3.73±0.81, P <0.05), ET group (2.35±1.05 vs 3.60±0.99, P <0.05) and ET+PH group (2.35±0.88 vs 3.4±0.85, P <0.01). Conclusion: The effect of ischemia on the Na +,K + ATPase activity is associated with different factors inducing ischemia; IND may influence the effect of ET on the Na +,K + ATPase activity.
