摘要
[目的]观察甲基莲心碱(Nef)增强顺氯氨铂(CDDP)对Lewis肺癌的化疗作用。[方法]建立小鼠Lewis肺癌皮下移植瘤模型,40只雄性荷瘤小鼠随机分成生理盐水(NS)、Nef、CDDP、Nef+CDDP四组分别给药,实验末采血并处死小鼠,检测外周血ALT、AST、BUN、CR,取皮下瘤块称重,计算瘤重抑制率,肉眼和镜下观察记录肺转移结节数,计算肺转移结节抑制率,金氏公式计算q值评价联合用药效应,石蜡切片镜下观察肿瘤组织的病理形态。[结果](1)NS、Nef、CDDP、Nef+CDDP组平均瘤重分别为(7.80±0.57)g、(6.27±1.31)g、(3.31±0.40)g、(1.55±0.25)g,平均肺转移结节数分别为(4.10±1.29)个、(3.90±0.99)个、(2.80±1.03)个、(1.00±0.82)个,CDDP和Nef+CDDP组平均瘤重和肺转移结节数明显低于NS组与Nef组(P<0.01或P<0.05)。(2)CDDP组和Nef+CDDP组的血清ALT、AST、BUN、CR值均明显高于NS组和Nef组(P<0.01),而CDDP组与Nef+CDDP组比较无显著性差异(P>0.05)。(3)镜下观察,CDDP组瘤细胞稀疏,可见瘤细胞变性、坏死;Nef+CDDP组较CDDP组瘤细胞变性、坏死更为明显。[结论]甲基莲心碱具有增强CDDP对Lewis肺癌的化疗作用。
To investigate the enhancing effect of Neferine in cisplatin chemotherapy for mice C57BL/6trans-planted with Lewis lung carcinoma.Forty male mice C57BL/6transplanted with Lewis lung carcinoma were randomly divided into4groups,(1)normality saline group,(2)Nef group,(3)CDDP group and(4)CDDP+Nef group.All mice were sacrificed at the end and blood was collected for biochemistry detection.Executed subcutaneous tumors were weighted to calculate the inhibitory rate of tumor weight.The metastatic nodus in lung were counted.The inhibitory rate of metastatic tumors in lung was measured with Jin’s formula.Paraffinslices were studied histopathologically.For NS,Nef,CDDP,and CDDP+Nef group,the mean weight of subcutaneous tumors were(7.80±0.57)g,(6.27±1.31)g,(3.31±0.40)g and(1.55±0.25)g respectively,and the mean metastatic nodus were4.10±1.29,3.90±0.99,2.80±1.03,1.00±0.82respectively.The tumor weight or number of nodus in group3and group4was significantly less than that in group1and group2(P<0.01,P<0.05).No significant difference in serum ALT,AST,BUN and CR was found between group3and group4(P<0.05).Serum ALT,AST,BUN and CR in group3or group4was significantly higher than those in group1or group2(P<0.01).Tumor cells sparseness degeneration and necrosis were found in group3with more seri-ous in group4.[Conclusion]Neferine can enhance chemotherapeutic effect of CDDP in the mice C57BL/6transplanted with Lewis lung carcinoma.
