摘要 目的 研究抗瘤新药PTS(抑瘤仙 )体外对肺癌细胞、耐药肺癌细胞及对正常支气管上皮细胞的作用 ,为其临床应用提供理论依据。方法 以大细胞肺癌株NCI H 460、耐药大细胞肺癌株H 460 /cDDP及正常支气管上皮细胞株 16HBE为实验对象 ,采用 2 4h活细胞跟踪法、MTT法及台盼蓝拒染记录细胞生长曲线法 ,进行PTS的体外抗瘤作用研究 ,同时以DDP(顺铂 )为阳性对照 ,PBS(平衡盐液 )为阴性对照。结果 PTS对受到直接损伤的肺癌细胞及耐药肺癌细胞均有杀伤作用 ,具有抗瘤作用和一定的抗耐药性。与DDP比较 ,PTS有更强的抗瘤作用 ,对于H 460细胞在第 48h 5 0 0mmol的DDP杀伤率为 62 .1% ,1/2 0 0的PTS为 81.3 % ;而PTS对于正常人支气管上皮 16HBE细胞的影响较小 (DDP为 3 9.5 % ,PTS为 2 1.6% ) ,具有较高的选择性。PTS应用后立即起效并达高峰 ,作用持续时间短。结论 PTS体外抗瘤作用明显 。 Objective To evaluate the effect of the new anti-cancer drug PTS(Para-Toluenesulfonamide Injection) on lung cancer cells,multi-drug resistance lung cancer cells and normal bronchial epithelium.Methods Human large cell lung cancer cell line NIH-H460,multi-drug resistance large cell lung cancer cell line H460/cDDP and normal bronchial epithelium cell line 16HBE were enrolled in the study.The anti-cancer effect in vitro was assessed by investigating the morphology of the alive cell in the following 24 h,MTT assay and the recording of growth curve by typan blue rejected staining.DDP(Cisplatin) was served as positive control and the balance salt buffer(PBS) as negative control.Results PTS possessed a stronger anti-cancer effect as compared with DDP.It also demonstrated the similar effect on drug-resistant H460/cDDP cells.In the H460 cell group,the killing rate by PTS(1/200) at 48 h was 81.3%,as compared with 62.1% by DDP.Less impact on the normal bronchial epithelium(16 HBE) was found in PTS,comparing with DDP(killing rate:21.6% vs 39.5%).It indicated the selective effect of PTS on cancer cell line.The anti-cancer effect appeared immediately after adding PTS,reaching the peak within one minute,remaining stable within 24 h.Conclusion PTS possesses selective and remarkable killing effect on H460 cells in vitro,and remains equally effective on DDP resistant H460 cells(H460/cDDP).
