摘要
目的建立赶黄草中槲皮素反相高效液相色谱法(RP-HPLC)含量测定方法。方法采用RP-HPLC对槲皮素进行定量测定;采用Kromasile C18柱(4.6 mm×150 mm,5μm)色谱柱,检测波长为256 nm。结果 色谱条件为:流动相:甲醇-0.2%磷酸溶液(55:50),柱温:25℃;流速:1.0mL·min-1。此法线性范围6.7~40.2μm·mL-1,r=0.9980,方法的精密度RSD为1.65%(n=5)。平均回收率为98.3%,RSD为1.73%(n=5)。结论 测定方法简便快捷、精密度高、准确性好,可以作为赶黄草中黄酮的测定方法,以控制其原料的质量。
OBJECTIVE: To establish a RP-HPLC method for the determination of quercetin in Penthorum chinense Pursh. METHODS: The content of quercetin was determinated by RP-HPLC. Kromasile C18 column (4.6 mm × 150 mm,5 μm) was used. The detection wave-length was set at 256 nm. RESULTS: The mobile phase consisted of methanol-0.2% phosphatic acid (55:50) with flow rate 1.0 mL·min-1 and column temperature at 25 °C. The method was liner within the range of 6.7 ∼ 40.2 μg·mL-1 (r = 0.9980). The method RSD was 1.65% (n = 5). The average recovery of assay was 98.3% and RSD was 1.73% (n = 5). CONCLUSION: The method appeared to be simple and rapid, highly accurate and precise. It can be used for the quantitative determination of quercetin in Penthorum chinense Pursh.
