摘要
目的 :建立RP HPLC法同时测定人血清中阿莫西林、克拉维酸钾的浓度。方法 :采用LunaC18色谱柱 (2 5 0mm× 4 .6mm ,5 μm) ,以 0 .0 5M磷酸盐缓冲液 乙腈 (90∶10 ;磷酸调pH至 2 .30 )为流动相 ,流速 1ml·min-1。检测波长为 2 2 0nm。结果 :CLV在 0 .0 6 6~ 16 .86 μg·ml-1,范围内有良好线性关系 ,r=0 .9999(n =9) ;AMO在 0 .12~ 2 9.4 5 μg·ml-1,范围内有良好线性关系r=0 .9998(n =9)。本法测定血清中CLV最低检测浓度为 0 .0 6 0 μg·ml-1,AMO为 0 .10 μg·ml-1。 (S/N≥ 3)。平均方法回收率为 97.5 %。结论 :本法简便 ,准确 ,重现性好 ,用于阿莫西林克拉维酸钾分散片人血清中血药浓度测定取得良好结果。
Objective: A simple RP-HPLC method was established for the determination of AMO & CLV in human serum.Method: The chromatographic separation was carried out on Luna C 18 column(250 mm× 4.6 mm, 5 μm) with a mobile phase consisting of 0.05M NaH 2PO 4 buffer-acetonitrile (90∶10, adjust pH to 2.3 with H 3PO 4), flow rate was 1 ml·min -1. The detecting wavelength was at 220 nm.Result: The linear range of CLV and AMO were 0.066- 16.86 μg·ml -1, 0.12- 29.45 μg·ml -1,respectively. The minimum detection limit of CLV and AMO were 0.060 μg·ml -1, 0.10 μg·ml -1(S/N=3),respectively;The average recover in two drugs rates were 97.5%. Conclusion: This method is simple, accurate, and reproducible. It has been applied successfully to the determination of AMO and CLV dispersible tablets in human serum for pharmacokinetic studies.
