摘要
目的:观察从室管膜前下区(anteriorsubventricularzone,SVZa)沿喙侧迁移流(rostralmigratorystream,RMS)至嗅球通路上碱性成纤维细胞生长因子(basicfibroblastgrowthfactor,bFGF)的表达情况。方法:应用地高辛标记的酶标原位杂交技术显示新生7d大鼠脑内室管膜前下区经迁移流至嗅球区域中bFGFmRNA的表达情况。结果:新生7d大鼠的脑内广泛表达bFGFmRNA,尤以SVZa,RMS和嗅球区域最为明显,其强度又以SVZa最强,阳性细胞数量最多,密度最大,嗅球最弱。在嗅球区阳性产物主要沉积在细胞质内,而在SVZa和RMS区则以核内较多。结论:bFGF对于维持SVZa区神经干细胞(neuralstemcell,NSC)的增殖状态密切相关,而NSC为了保持自身的增殖状态则表达了较高含量的bFGF。在SVZa区bFGF主要以促进细胞增殖为主,而在嗅球区bFGF则以促进细胞分化为主。
AIM: To observe the expression of basic fibroblast factor(bFGF)in the pathway from anterior subventricular zone(SVZa)through rostral migratory stream(RMS)to olfactory bulb. METHODS:The mRNA expression of bFGF in the brain of newly born 7-day rat wa s measured by the digoxin-labeled in situ hybridization. RESULTS:The message RNA of bFGF was expressed extensively in the brain of new ly born rat(aged 7 days), and the most obviously expression regions were located in the SVZa,RMS and olfactory bulb.The intensity of the positve reactivity was the strongest in SVZa and the weakest in olfactory bulb.The number of the posit ive cells in SVZa was the most and the density of the positive cells was also th e highest.The positive products mainly deposited in the cytoplasm of olfactory b ulb,but the positive products mainly deposited in the nucleus of SVZa and RMS. CONCLUSION:bFGF is closely related to maintaining the proliferation status of neural stem cells(NSC)in SVZa.To maintain the proliferation,NSC also expresses more quantity of bFGF.bFGF mainly accelerates the proliferation of the cells in SVZa,while mainly stimulates the differentiation of the cells in the olfactory b ulb.
