摘要 目的:观察长期脑缺血再灌注大鼠脑的形态和组织病理学的变化,探讨建立脑萎缩模型的可行性。方法:再灌注后结扎双侧颈总动脉,动态监测大鼠缺血后行为学、脑质量,同时苏木精-伊红染色观察脑的组织病理学变化。结果:大鼠脑缺血后脑萎缩出现于2~4个月,随时间延长而加重,对照组脑质量:4个月时为(2.080±0.107)g,6个月时为(2.213±0.072)g,脑缺血组大鼠脑质量逐渐减轻,6个月降至(1.834±0.059)g,较对照组差异有非常显著性意义(t=4.287,P<0.01)。苏木精-伊红染色显示脑组织变性坏死,细胞丢失严重。结论:再灌注后结扎双侧颈总动脉可建立较稳定的脑萎缩模型,较好的模拟了临床上反复脑缺血导致的脑萎缩的发展过程。 AIM:To observe morphologic and histopathologic change of rat brains with long-term cerebral ischemia reperfusion and to discuss the possibility of the mode l of brain atrophy. METHODS:After reperfusion the common carotid arteries of the rats were ligate d.Dynamic observation was conducted in behavior and brain weight of the ischemic rats.Meanwhile,HE staining was used to observe the histopathological changes of the rat brains. RESULTS:Brain atrophy, which was aggravated as time went by,appeared 2 to 4 m onths after cerebral ischemia.The average weight of the ischemic rat brain reduc ed significantly to(1.834± 0.059) g at 6 months after ischemia,as compared with that of the control rats [(2.080± 0.107) g at 4 months after ischemia and (2.2 13± 0.072)g at 6 months after ischemia](t=4.287,P< 0.01).HE staining showed tha t the brain had degeneration and necrosis with severe loss of neurons. CONCLUSION:The model of brain atrophy can be steadily established by the comm on carotid artery occlusion,which preferably simulates the development of brain atrophy caused by repetitive cerebral ischemia in clinic.
