摘要
目的:探讨重组人骨形成蛋白成熟肽-2(recombinanthumanbonemor-phogeneticprotein-2mrhBMP-2m)对辐射引起的小鼠骨髓造血损伤的修复作用。方法:通过60Coγ射线照射小鼠,建立骨髓造血损伤动物模型;经rhBMP-2m连续治疗后,比较照射对照组和rhBMP-2m治疗组之间,骨髓单个核细胞数和蛋白含量的差异;同时,通过酶联免疫吸附试验(ELISA),检测骨髓细胞中CD34分子表达水平的变化。结果:经rhBMP-2m治疗后的动物,其单个核细胞数、蛋白含量和CD34分子的表达水平明显高于照射对照组(P<0.05,n=5)。结论:辐射引起的小鼠骨髓造血损伤,rhBMP-2m能够增加造血细胞数量,促进骨髓细胞CD34分子表达,加速造血损伤的修复过程。
AIM:To discuss the repair effect of recombinant human bone morphogenetic protein 2m(rhBMP 2m) on bone marrow hematopoietic injury induced by irradiation in mice. METHODS:The animal model of bone marrow hematopoietic injury was established through radiating the mouse with 60Co γ ray.After continuous treatment with rhBMP 2m,the differences in the number of mononuclear cells and the concentration of protein were compared,and meanwhile the variance of CD34 molecule was detected by the method of enzyme linked immunoadsordent assay(ELISA),both between the control group and the reBMP 2m treatment group. RESULTS:The expression levels of CD34 molecule,the number of mononuclear cells and the concentration of protein and the variance of the treatment group were all higher than those of the control group(P< 0.05,n=5). CONCLUSION:rhBMP 2m can increase the number of mononuclear cells,promote the expression of CD34 and accelerate the repair process of hematopoietic injury in mice with the bone marrow hematopoietic injury induced by irradiation.
