摘要 目的:探讨辣椒素对增生性瘢痕成纤维细胞增殖能力和胶原合成的影响,以期为辣椒素在瘢痕防治的临床应用方面提供实验依据。方法:取增生性瘢痕10例,采用组织块培养法获得原代成纤维细胞,选用4代细胞为实验模型,将辣椒素(2~100mg/L)加入细胞培养体系中进行干预并与对照组比较,用3H-TdR和3H-脯氨酸参入法分别检测成纤维细胞增殖能力和胶原合成水平的变化。结果:辣椒素对成纤维细胞的作用表现为剂量和时间依赖性。高含量的辣椒素(>50mg/L)对成纤维细胞有直接的损伤作用,作用24h后可导致细胞死亡;2和10mg/L低含量的辣椒素在不杀伤细胞的同时,能够显著抑制增生性瘢痕成纤维细胞3H-TdR和3H-脯氨酸参入,仅为未用药组的50%和30%。结论:辣椒素可以导致培养的增生性成纤维细胞功能发生明显改变,细胞增殖能力和胶原合成能力受到显著抑制。结合同期临床观察表明,辣椒素在瘢痕防治的临床应用方面具有潜在的价值。 AIM:To investigate the effect of capsaicin on the proliferation and collagen synthesis of hypertrophic scar derived fibroblast in order to provide an experimental basis for the application of capsaicin in the clinical prevention and treatment of scar. METHODS:Hypertrophic scar derived human fibroblasts in 10 patients were cultured in vitro by tissue block culture. Four generation cells were selected as the experimental models.The capsaicin(2-100 mg) was added to the culture media of the cells,intervention was performed,and then were compared with the control group.The changes in the levels of the collagen synthesis and proliferation of the fibroblasts were examined by the incorporation values of 3H TdR and 3H proline into the cells. RESULTS:The effects of capsaicin on the fibroblasts represented were represented by the dependence on the dose and time.Capsaicin could directly damage the fibroblasts at high concentration(≥ 50 mg/L) and cause the death of cells when capsaicin was applied more than 24 h.Capsaicin at low doses of 2 and 10 mg/L did not kill the cells as well as significantly inhibited participation of the hypertrophic scar derived fibroblast 3H TdR and 3H proline,which were only 50% and 30% of those in the control group respectively. CONCLUSION:Capsaicin can change the function of hypertrophic fibroblasts,and directly inhibit the proliferation of fibroblasts and collagen synthesis. It may be a potential agent for the prevention and treatment of hypertrophic scar.
