摘要
目的 探讨细胞周期素D 1、E(cyclinD1、cyclinE)、p2 1cip1在pTNMⅠ期普通型肾细胞癌 (CRCC )发生发展中的作用。方法 用半定量RT PCR方法 ,检测 2 3例 pTNMⅠ期CRCC组织和 7例正常肾组织中cyclinD 1、cyclinE及 p2 1cip1的mR NA含量 ;用免疫组化方法检测CRCC中 p5 3及ki 67的表达 ;并将以上指标与临床病理特征进行统计分析。 结果 CRCC组织中cyclinD1、cyclinE、p2 1cip1mRNA含量分别为 0 .484± 0 .416、0 .472± 0 .3 40、0 .3 5 6± 0 .2 15 ,均高于正常对照组 ( 0 .0 78± 0 .0 5 1,P <0 .0 1;0 .0 63± 0 .0 63 ,P <0 .0 1;0 .178± 0 .114 ,P <0 .0 5 ) ;CyclinD1含量低者肿瘤直径小于 3cm (P <0 .0 1) ;CyclinE含量低者肿瘤常有坏死 (P <0 .0 5 ) ,细胞核分级较高 (P <0 .0 1) ;p2 1cip1低表达者细胞核分级较高 (P <0 .0 5 ) ;p2 1cip1与cyclinE呈正相关 (γ =0 .60 2 ,P <0 .0 1)。结论 pTNMⅠ期CRCC组织中cyclinD1、cyclinE、p2 1cip1mRNA含量升高 。
Objective To investigate the role of cyclin D1?cyclin E ?p21 cip1 in the carcinogenesis and development of pTNM stage Ⅰ CRCC.Methods The mRNA expression of cyclin D1 ?cyclin E and p21 cip1 was detected by RT-PCR in 23 pTNM stage Ⅰ CRCCs and 7 normal renal tissue distal to tumor.Immunohistochemistry was used to investigate the expression of p53 and ki-67 in pathological tissue sections of CRCCs.The relationship between those index and clinicopathological parameters were analyzed statistically.Results The mRNA expression of cyclin D1?cyclin E and p21 cip1 were significantly different between CRCC and normal control(0.484±0.416 vs 0.078±0.051,P<0.01?0.472±0.340 vs 0.063±0.063,P<0.01 and 0.356±0.215 vs 0.178±0.114,P<0.05).The tumor with a low cyclin D1 had a size less than 3 cm(P<0.01).The lower expression of cyclin E was related to tumor necrosis(P<0.05) and high nuclear grade(P<0.01).The lower expression of p21 cip1 was also related to high nuclear grade(P< 0.05).There was positive correlation between p21 cip1 and cyclin E(γ=0.602,P<0.01).Conclusion The mRNA expression of cyclin D1?cyclin E and p21 cip1 are all higher in pTNM stage Ⅰ CRCC than in the normal control tissue.The increased expression may affect the biologic action and prognosis of the tumors.
