摘要
以黄杆菌(Flavobacterium)对硫磷水解酶(parathionhydrolase)的序列(AAA24930.1)对芽孢杆菌的基因组进行BLASTP,从Bacillushalodurans中发现两个对硫磷水解酶类似蛋白PTEa和PTEb,长度分别为679和331个氨基酸。利用conserveddomainsearch和motifscan软件分析的结果证实它们具有对硫磷水解酶的结构特征。PTEa和PTEb的对硫磷水解酶上游标志区分别是"GVCACHEHL"和"GFTYSHEHI",与标准的"G-x-T-L-x-H-E-H-[LIV]"大致吻合。PTEb的对硫磷水解酶下游标志区"AVARAHHETKAPIHSH"也与典型的"A-x-A-x-A-x(4)-G-x-P-[LIVM]-x(2)-H"基本一致,而PTEa中缺乏明显的下游标志区。PTEb序列对枯草杆菌基因组TBLASTN的结果表明枯草杆菌也可能存在对硫磷水解酶类似蛋白。
Two putative parathion hydrolases of Bacillus halodurans, PTEa with 679 amino acid residues and PTEb with 331 amino acid residues, were found by using the protein sequence of parathion hydrolase from Flavobacterium to BLASTP the genomes of 4 Bacillus strains. Results from conserved domain search and motif scan further demonstrated that the PTEa and PTEb were indeed parathion hydrolase. The upstream consensus patterns were 'GVCACHEHL' in PTEa and 'GFTYSHEHI' in PTEb against the typical 'G-x-T-L-x-H-E-H-[LIV]'. And 'AVARAHHETKAPIHSH' was the downstream consensus pattern found in PTEb against the typical 'A-x-A-x-A-x(4)-G-x-P-[LIVM]-x(2)-H', while no obvious consensus pattern could be found in the downstream of PTEa. The TBLASTN results of the PTEb sequence against the genome of Bacillus subtilis subsp. subtilis str. 168 showed that parathion hydrolase might also be encoded by the genome of Bacillus subtilis.
