摘要
以结缕草愈伤组织为试材,利用农杆菌介导法时包括辐照、愈伤组织状态、共培养条件及负压处理等辅助处理方法对愈伤组织瞬间表达频率的影响进行研究。结果表明:2Gy的辐照剂量利于提高瞬间表达频率;1~2个月的愈伤组织适于转化;全光照的共培养条件下瞬间表达频率高于16/8h光照培养和暗培养条件下瞬间表达频率;农杆菌与愈伤组织混合培养时给予适当的负压条件可提高愈伤组织瞬间表达频率和存活率。
The assistant method of agrobacterium-mediated transformable efficiency includes irradiation, callus, co-cultivation and negative pressure, was explored with the callus of Japanese Lawngrass(Zoysia japonica Steud.). The procedure was as follows: 2 Gy of irradiation-dosage benefited the exaltation of transformable efficiency; 1~2 month of callus was fitter to the transformation. The whole light shine to co-cultivation had higher transformation efficiency than the 16/8 h photoperiod and of the whole dark. proper negative pressure could increase transformation frequency and survival rate of callus when agrobacterium and callus co-cultivated.
