摘要
目的研究与张力蛋白和辅助蛋白同源、第10号染色体丢失的磷酸酶基因(PTEN)蛋白和细胞周期素D1(CyclinD1)在膀胱移行细胞癌(TCC)中的表达,探讨其与TCC生物学行为的关系。方法应用免疫组织化学方法观察56例TCC标本PTEN蛋白和CyclinD1的表达情况。结果(1)TCC组中PTEN蛋白阳性率为46%,明显低于正常膀胱黏膜组(P<0.01);CyclinD1阳性率为53%,明显高于正常膀胱黏膜组(P<0.01)。随着肿瘤恶性程度的增高和临床分期的进展,PTEN蛋白阳性率下降,CycliD1阳性率上升。(2)PTEN蛋白和CyclinD1的表达呈显著负相关(P<0.01)。(3)有32%的病例存在PTEN蛋白和CyclinD1同时表达或失表达。结论PTEN蛋白和CyclinD1的异常表达在TCC的发生、发展过程中起重要作用。两者同时检测有助于预后判断。
Objective To investigate the expression of PTEN protein and CyclinD 1 in transitional-cell carcinoma of the bladder and to evaluate its clinical significance.Methods The expression of PTEN protein and CyclinD 1 were detected with immunohistochemistry in56cases of transitional-cell carcinoma (TCC).Re -sults The positive rate of PTEN protein in TCC were lower than that in normal bladder mucosa(46%vs100%,P<0.01);while the positive rate of CyclinD 1 expression in TCC was higher than that in normal bladder mucosa(53%vs16%,P<0.01).With the progress of the tumor grade and clinical stage,the positive rate of PTEN protein in TCC decreased;while that of CyclinD 1 increased.PTEN protein expression was negatively correlated with CyclinD 1 expression.Dual-positive and dual-negative expression of two proteins occurred in32%of all cases.Conclusion Results suggest that the abnormal expression of PTEN and CyclinD 1 may play an important roles in the carcinogenesis and progression of TCC.Detection of both proteins might be helpful in prognosis of TCC.[
