摘要
目的 :体外诱导鼠骨髓间充质干细胞 ,与聚乳酸 -聚羟基乙酸支架材料体外复合构建人工骨进行研究。方法 :分离培养大鼠骨髓间充质干细胞 ,并进行体外诱导和成骨能力检验 ;而后与聚乳酸 -聚羟基乙酸支架材料复合构建三维培养体系 ,进行扫描电镜、碱性磷酸酶和骨钙素活性检测。结果 :对已构建三维培养体系 ,扫描电镜检测见有大量细胞粘附生长 ;均检测到碱性磷酸酶和骨钙素活性。结论 :已构建三维培养体系内鼠骨髓间充质干细胞仍具有良好的增殖能力和成骨活性 。
Objective:To study the induction of mesenchymal stem cells(MSCs) derived from rat bone marrow in vitro and the construction of artificial bone by implanted MSCs into the Polylactic-Polyglycolic acid(PLGA) cradle.Methods:MSCs were isolated from rat bone marrow and cultured with DMEM, then induced and detected the ability of bone formation. The three-dimensional system was constructed by implantation of MSCs into PLGA cradle, detected by scanning electron microscope, and measured the activity of ALP and Osteocalcin.Results:To the three-dimensional system constructed , the detecting results showed that there were many cells on the surface of the cradle in the photographs developed by scanning electron microscope, and that MSCs possesed the activity of ALP and Osteocalcin.Conclusions:MSCs still possess the ability of proliferation and bone formation in the three-dimensional system, and they may be used to construct artificial bone.
