摘要
目的 探讨新型抗心肌缺血药———果糖二磷酸镁 (FDP Mg)的安全性。 方法 采用Bliss法和QUANTAL程序测定和计算了FDP Mg的LD50 和 95 %可信限并对其过敏性、溶血性和血管刺激性进行研究。结果 FDP Mg小鼠静脉注射的LD50 为 2 84 .81mg/kg ,95 %可使限为 2 6 7.4 2~ 30 2 .2 0mg/kg ;FDP Mg大鼠静脉注射的LD50 为 335 .15mg/kg ,95 %可信限为315 .75~ 35 4 .5 5mg/kg ;FDP Mg大鼠静脉滴注 [10 0 0mg/ (kg·h) ]的MLD为 (2 5 0 9.9± 2 2 6 .8)mg/kg。FDP Mg不引起全身过敏反应和溶血现象 ,对血管也无刺激性。结论 FDP Mg用于静脉给药安全可靠。
OBJECTIVE To test security of magnesium fructose-1,6-diphosphate (FDP-Mg), a novel drug of anti-myocardial ischemia. METHOD Bliss method(QUANTAL procedure)was used to calculate LD 50 and 95% confident limit. Allergic, hemolytic and irritant tests of blood vessel were done.RESULTS LD 50 of FDP-Mg for mice through intravenous injection was 284.81mg/kg and 95% confident limit was 267.42~302.20mg/kg, LD 50 of FDP-Mg for rats through iv was 335.15mg/kg and 95% confident limit was 315.75~354.55mg/kg and MLD of FDP-Mg for rats through intravenous drip by [1 000mg/(kg·h)] was (2 509.9±226.8)mg/kg. FDP-Mg did not induce general allergic reaction and hemolysis, the irritant test of blood vessel was negative. CONCLUSION It is safe to administrate FDP-Mg by intravenous drip.
