摘要
目的:检测雄激素及其受体对maspin基因启动子的作用。方法:构建含maspin基因5侧启动子区846-759+87bpDNA的荧光素酶报告基因表达载体pGL3-mP,然后与雄激素受体表达载体hAR共转染雄激素非依赖型前列腺癌细胞株PC-3M,用或不用雄激素处理细胞,48h后应用双荧光素酶测定系统,检测荧光素酶的表达活性。结果:hAR可抑制maspin启动子的表达活性,其抑制作用是雄激素配体非依赖性的。结论:maspin启动子区可能含有与雄激素受体结合的负性调控元件。
Objective: To detect the effect of androgen and its receptor on the promoter of maspin. Methods: pGL3-mP luciferase expression vector containing 846bp(-759,+87) DNA of maspin gene 5'promoter region was constructed. Then pGL3-mP plasmid and androgen receptor expression vecetor were cotransfected into PC3-M cell treated with or without androgen. Through detecting the activity of luciferase, the effect of androgen and its receptor on the activity of maspin promoter was studied. Results: Androgen receptor suppressed the activity of maspin promoter and its effect was independent from androgen. Conclusion: It is possible that the region of maspin promoter contains negative regulatory elements combined with androgen receptor.
